Indications & Dose:
Dietary supplement, vitamin D deficiency, refractory rickets, renal osteodystrophy, hypoparathyroidism, hypophosphatemia, hypocalcemic tetany, osteoporosis
BY INJECTION
Rickets:
1 ampoule of 200000 i.u of D3 every six months or 2 ampoules of 400000 i.u every six months if child is rarely exposed to sun or heavily pigmented.
Osteomalacia:
Same as rickets.
Orally or IM inj: Adult ; 10mg twice a year (5mg per dose)
Antenatal prophylaxis:
1 ampoule of at the start of 7th month of pregnancy.
Lactation
ampoule of at the start of breast feedign taking into accoutn any previous administration with a minimum interval of 4 months.
Prevention of vitamin D deficiency
Adult: 400 units daily
Treatment of vitamin D deficiency
Adult: 800 units daily, higher doses may be necessary for severe deficiency
Contraindications:
Hypercalcemia, renal dysfunction, hyperphosphatemia, abnormal sensitivity ‘to the toxic effects of vitamin D and hypervitaminosis D
Side Effects:
CNS: Drowsiness, fatigue, headache, psychosis, seizures, weakness, anorexia
CV: Dysrhythmias, hypertension, vascular calcification
GI: Constipation, cramps, diarrhea, dry mouth, metallic taste, nausea, vomiting, pancreatitis, elevated ALT/AST
GU: Albuminuria, decreased libido, hematuria, nocturia, polyuria, reversible azotemia
METAB: Hypercholesterolemia, mild acidosis
MS: Bone pain; muscle pain, weakness
SKIN: Photopfiobia, pruritus
MISC: Weight, loss

(400 IU/d); (doses above recommended daily allowance) associated with supravalvular aortic stenosis, elfin facies, and mental retardation; caution should be exercised when ergocalciferol is administered to nursing women; vitamin D and metabolites appear in breast milk; compatible with breast feeding, but infant should be monitored for hypercalcemia if doses exceed recommended daily allowance
