Name: Polymyxin B Sulphate

Indications & Dose:

It is active against gram-ve organism including pseudomonas aero-ginosa skin infections.
DOSE
Adults: 0.25-1gm four times 
"Children: 12.5mg/kg/dose . Oral: in diarrhoea and 3g/M2/day q 6 hr. oral in hepatic coma

Precautions:

Renal dysfunction, perforated tympanic membrane. When given i.m. or intrathecally, administer only to hospitalised patients.


Pregnancy & breast-feeding: Safety not established.


Old age: Use with caution. Large open wounds.

Drugs


Aminoglycosides: Concurrent use may increase risk of respiratory paralysis and renal dysfunction.


Nondepolarising muscle relaxants: Neuromuscular blockade may be enhanced.


Loop diuretics: The auditory toxicity appears to be increased.


Anaesthetics, Neuromuscular blocking agents (Tubocurarine, Gallamine): Risk of neuromuscular blockade and respiratory paralysis.


Penicillin V, Methotrexate, 5-fluorouracil, oral Vit. B12, Digoxin: Oral neomycin inhibits the GI absorption of these drugs.


Coumarin: Oral, neomycin enhances effect of coumarin
